Have you tried plugging it in to a different USB port. Some ports are no connected directly to the Hub and so can cause problems to start with (though, once a connection is established, they are usually fine). Try a different USB port first and see what happens.
Does the PC make a "Chime" sound a few seconds after plugging the phone in? If so, it is connecting and the problem could just be that WMDC (Windows Mobile Device Centre) isn't picking it up.
